Hello I was wondering what vacuum port should i use for the brakes . I put a new intake and carb on . The carb has a brake vacuum and so does the manifold . Does it really made a difference on what one i use ? Thanks again

As long as you have full manifold vacuum to the booster, you're fine. Doesn't matter which one you use.

What should the amount of vacuum be ?? Is there a set or appropiate number with a vacuum gauge?

Yeah you should be making 18-20 in-hg on a stock engine.
